The 12C was the first series-production road car from the modern McLaren Automotive and established the carbon-fiber MonoCell, twin-turbo V8 and hydraulic ProActive Chassis Control formula used by several later McLaren models. It directly competed with cars such as the Ferrari 458 Italia and laid the foundations for the 650S. It was offered as either a coupé or Spider, with either variant offering identical levels of performance.

The 12C HS was one the earliest creations from McLaren Special Operations, turning the standard 12C into a rarer and more aggressive road car. Along with increased engine output, it received GT3-inspired aerodynamic bodywork, including revised bumpers, larger vents, a diffuser and a more prominent rear wing.

The 12C GT Sprint is a track-only bridge between the road-going 12C and full 12C GT3 race car. It combines the road car's full engine output with GT3-inspired aerodynamics, racing slicks, a 40mm lower ride height, race brakes, roll cage and air-jack system, while retaining systems such as ProActive Chassis Control and the active Airbrake.

The 12C GT Can-Am Edition is an unrestricted, track-only evolution of McLaren's GT3 racer, created as an extreme private track car rather than a competition machine. It combined increased power, 1200kg of dry weight and much more aggressive aerodynamics with slick tires and fully racing equipment, making it one of the rarest and most focused derivatives of the 12C.

The 12C GT3 marked McLaren Automotive's return to customer sports-car racing and achieved 19 race victories during its 2012 debut season. It replaced the road car's dual-clutch gearbox with sequential racing transmission and added race-specific aerodynamics, suspension, brakes, and safety equipment, before eventually being succedded by the 650S GT3.
